LaToya Johnson and Asia Taylor both had 10 points for the Cardinals (1-0), who trailed 60-53 with 4:53 to play.
Kendel Ross had 17 points and nine rebounds for the Flyers (1-1), who opened the season with a victory over No. 10 Michigan State.
The Cardinals used an 8-1 run to tie the game at 61 with 2:25 to go.
Ross made two free throws with 37 seconds left to give the Flyers a 63-61 lead. Becky Burke's 3-pointer with 18 seconds to play gave Louisville a 64-63 lead.
